使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(上圖)或 β-Actin (D6A8) Rabbit mAb #8457(下圖)對未經處理 (-) 或經 λ 磷酸酶處理 (+) 的 RAW 264.7 和 Neuro-2a 細胞的提取物進行蛋白質印跡分析。
對 RAW 264.7 細胞提取物中的骨橋蛋白/SPP1 蛋白進行免疫沉澱分析。泳道 1 為 10% 輸入,泳道 2 為 Rabbit (DA1E) mAb IgG XP® Isotype Control #3900,泳道 3 為 Osteopontin/SPP1 (E9Z1D) Rabbit mAb。使用 Osteopontin/SPP1 (E4O2F) Rabbit mAb #27927 進行蛋白質印跡分析。Mouse Anti-Rabbit IgG (Light-Chain Specific) (D4W3E) mAb (HRP Conjugate) #93702 用作二抗。

使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb 對石蠟包埋的小鼠胰腺組織進行免疫組織化學分析。
使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb 對石蠟包埋的小鼠腦的腦橋區組織進行免疫組織化學分析。
使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(頂部圖)或 Osteopontin/SPP1 Rabbit mAb(底部圖)對石蠟包埋的 renca 同基因腫瘤組織(左圖)、4T1 同基因腫瘤組織(中圖)或小鼠肝臟組織(右圖)進行免疫組織化學分析。這兩種抗體均可檢測小鼠骨橋蛋白/SPP1 上的獨特、非重疊表位。使用兩種抗體獲得的相似染色模式有助於確認染色特異性。
與濃度匹配的 Rabbit (DA1E) mAb IgG XP® Isotype Control #3900(右圖)比較,使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(左圖)對石蠟包埋的小鼠腎臟組織進行免疫組織化學分析。
使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb 對石蠟包埋的 RAW 264.7 細胞沉澱物(左圖,陽性)或 Neuro-2a 細胞沉澱物(右圖,陰性)進行免疫組織化學分析。
對用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(左圖,綠色)標記的固定冷凍小鼠腎臟組織進行共聚焦免疫螢光分析。隨後將游離的二抗結合位點用 Rabbit (DA1E) mAb IgG XP® Isotype Control #3900 封閉,之後用 Ras (E4K9L) Rabbit mAb (Alexa Fluor® 488 Conjugate) #51813(右圖,紅色)和 DAPI #4083(右圖,藍色)進行共標記。
對用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(左圖,綠色)標記的固定冷凍小鼠丘腦組織進行共聚焦免疫螢光分析。然後將游離二抗結合位點用 Rabbit (DA1E) mAb IgG XP® Isotype Control #3900 封閉,之後用 Iba1/AIF-1 (E4O4W) XP® Rabbit mAb (Alexa Fluor® 647 Conjugate) #78060(右圖,紅色)和 DAPI #4083(右圖,藍色)共標記。
使用 Osteopontin/SPP1 (E9Z1D) Rabbit mAb(綠色)和 DAPI #4083(藍色)對 RAW 264.7 細胞(左圖,陽性)和 Neuro-2a 細胞(右圖,陰性)進行共聚焦免疫螢光分析。用 Brefeldin A #9972(100 ng/mL,90 分鐘)處理細胞以抑制蛋白質分泌。

Osteopontin/SPP1兔單克隆抗體用於識別內源水平的骨橋蛋白/SPP1 總蛋白。骨橋蛋白是一種在多種細胞類型中表達的高度磷酸化分泌型糖蛋白。分泌型磷蛋白 1 (SPP1) 基因編碼的骨橋蛋白最初從大鼠骨肉瘤細胞系中分離並克隆 【1】。骨橋蛋白是一種高度帶負電荷的蛋白質,該蛋白質可能調節骨骼和其他組織中的胞外基質周轉和重塑 【2】。骨橋蛋白還具有三個高度保守的受體結合結構域,也稱為精氨酸-甘氨酸-天冬氨酸 (RGD) 結構域。
